L27Rα (IL27R), also well known as WSX-1 and TCCR, is a type I cytokine receptor that functions only as the ligand binding component of the receptor for IL27, and together with the glycoprotein 130 (gp130) coreceptor constitutes a functional signal-transducing receptor mediating the IL27 effects. Neither receptor chain alone is sufficient independent of the other. As a member of the IL6/IL12 family of cytokines, IL27 is not only involved in the development of Th1-type adaptive immune response, but also acts on cells of the innate immune system. In addition, it has been demonstrated that IL27Rα, the single component of a heterodimeric cytokine receptor, can aberrantly activate signaling pathways and transform hematopoietic cells in a JAK-dependent manner in acute myeloid leukemia (AML). Accordingly, IL27Rα may play an unappreciated role in various myeloproliferative disorders (MPDs) and be identified as a future therapeutic target for this disease
